Programa de Genómica Funcional de Eucariotes

Este programa de investigación estudia a las PLANTAS, cómo modelo representativo de los sistemas eucariotes, analizando interacciones entre los vegetales y los microorganismos. Incluye tres grupos de investigación liderados por los investigadores: Dra. Georgina Hernández, Dr. Mario Serrano y Dr. Alexandre Tromas

Dos líneas de investigación del programa estudian la simbiosis leguminosa-rhizobia. Plantas de la familia de las leguminosas han desarrollado la capacidad de crecer en suelos privados de nitrógeno mediante la interacción con bacterias del suelo conocidas como rhizobia. Los 2 socios han co-evolucionado hacia una interacción muy compleja y positiva para ambos, en la que se lleva a cabo la fijación simbiótica de nitrógeno para capturar el nitrógeno atmosférico directamente y así mantener el crecimiento vegetal.

La línea de investigación del grupo de Georgina Hernández / Mario Ramírez es la genómica funcional de frijol (Phaseolus vulgaris) para el análisis de la simbiosis con Rhizobium etli y las respuestas de la planta a estreses abióticos. El frijol es la leguminosa más importante para el consumo humano en el mundo. Su secuencia genómica se publicó recientemente ( El grupo realiza investigación post-genómica de frijol, utilizando principalmente la transcriptómica, tanto de mRNAs que codifican para proteínas como de RNA pequeños no-codificantes identificados como reguladores esenciales. Uno de sus objetivos es describir nuevos reguladores globales de la simbiois frijol-rhizobia: factores de transcripción y microRNAs. Otro de los objetivos es el análisis transcriptómico del frijol en respuesta a estreses abióticos como la deficiencia de nutrientes, el estrés oxidativo y la sequía.

La línea de investigación del grupo de Alexandre Tromas es la regulación del citoesqueleto durante la fijación simbiótica de nitrógeno.Se ha establecido que el acomodo de los microsimbiontes, como rhizobia, dentro de la célula vegetal es un proceso activo que requiere del adecuado re-arreglo del citoesqueleto. Las Rho-GTPasas (denominadas ROP, en las plantas), consideradas como “switches moleculares”, activan el re-arreglo de la actina, uno de los principales componentes del citoesqueleto. La pregunta actual es entender cuáles de los complejos para re-arreglo del citoesqueleto están involucrados en el acomodo de las bacterias simbióticas y entender cuando y donde se re-localizan y activan en los pelos radicales de Lotus japonicus. Para responder esta pregunta se desarrollarán un conjunto de biosensores que permitirían la observación a alta resolución de la activación de cada ROP en los pelos radicales, durante la simbiosis. Los objetivos de esta línea son: Definir el Interactoma de las proteínas ROP (biología molecular y bioquímica); Desarrollar biosensores de la activación de las proteínas ROP en respuesta a rhizobia (biología molecular y celular); Estudiar el fenotipo de mutantes de las proteínas ROP (fisiología vegetal).

La línea de investigación del grupo de Mario Serrano, es la caracterización de la inmunidad innata a Botrytis cinerea, El hongo necrótrofo Botrytis cinerea, conocido comúnmente como moho gris, afectan múltiples órganos de más de 200 especies vegetales, por lo que es considerado el segundo hongo fitopatógeno más dañino para la agricultura. Las primeras interacciones entre las plantas y los hongos tienen lugar en la epidermis, donde la planta no solo tiene la primera barrera física, que incluye la cutícula en los órganos aéreos, sino es donde los organismos se reconocen mutuamente y potencialmente la inmunidad innata vegetal es inducida. Sin embargo, los eventos tempranos que tienen lugar durante la infección por Botrytis cinerea y en particular la comunicación entre la planta y el patógeno en la cutícula vegetal, han sido poco explorados. Cabe resaltar, que mutantes en la síntesis de la cutícula en Arabidopsis thaliana y Solanum lycopersicum, presentan un “síndrome” relacionado con la interacción planta-patógeno, que incluye un incremento en la permeabilidad foliar, la acumulación de especies reactivas de oxígeno y una resistencia en contra de B. cinerea. El proyecto se basa en la identificación y caracterización de los elementos moleculares que vinculan la degradación de la cutícula y la inducción de la inmunidad innata de las plantas. Para lograr este objetivo, utilizamos el modelo de interacción Arabidopsis thaliana-Botrytis cinerea y las herramientas de genómica tradicional y genómica química.

Alternative splicing in common bean

The post-genomic era lead to the discovery of alternative splicing in eukaryotes (plants and animals). Splicing of introns from a pre-mRNAs transcribed from a single eukaryotic gene can be regulated by different factors and so generate alternative spliced transcripts or isoforms. The analysis of the great complexity at the transcriptional level generated by alternative splicing in legumes has only begun. Based in our common bean transcriptome data we are analyzing the alternative splicing in this legume (collaboration with S. Jackson, U. Georgia).

MicroRNAs as global regulators of common bean symbiosis and of the response to abiotic stress

Micro-RNAs (miRNAs), a class of small non-coding RNAs 21-24 nt long, are essential post-transcriptional regulators for plant development and response to stress; their characterization and function in legumes is yet scant. We are interested in enhancing the knowledge about miRNAs in common bean, focusing in SNF nodules. Projects in this area include: a) miRNAs expression profiles through RNA-seq and b) defining specific functions of selected miRNAs in nodule development and/or in the response of SNF common bean plants to abiotic stress using reverse genetic approaches.

Symbiotic transcriptome (common bean / rhizobium) of the response to abiotic stress

The effectiveness of the atmospheric N-fixation process occurring in the legume-rhizobia symbiosis (symbiotic nitrogen fixation = SNF) is highly dependent on the adequate gene expression both in the macro- and micro-symbiont. One of our research interests is to perform combined transcriptome analyses of the Rhizobium etli bacteroids and the common bean nodule cells to generate integral knowledge of global gene expression and regulation in this symbiosis. Such project is done in collaboration with the group of L.

Our group has participated in pioneer research on common bean transcriptome. This work has been done in collaboration with Carroll P. Vance, U. Minnesota – USDA, and other colleagues.
